If you are just trying to run a 16bit app on a 64bit Win OS. If its an MS-DOS application you can try DOSBox. If not then your options for running on windows are to install a 32bit version of windows, as already noted this can be accomplished with a virtual machine, or as a dual-boot. If you have Win 7 Pro or Ultimate look up "XP Mode". If not look at VirtualBox or VMware Player, you will need a copy of a 32bit version of windows and any 32bit version will work. Another option is to use Wine for Linux, again you would have to install a Linux operating system, and this again can be done either in a VM or as a dual-boot. One nice thing about Wine is that it will run 16bit windows programs in a 64bit Linux. Interfacing with hardware can get tricky when running a VM in Windows.
I have a 5 year old dell laptop running a 2.0GHz Intel Core2Duo (dual core) with 2GB of ram and it handles both XP mode and Linux on VirtualBox just fine, there is a little lag but not enough to be an annoyance. On the other hand my newer Asus netbook running a 1.8 Intel Atom (dual core) with 2GB of ram runs just a little on the slow side.
I will note that if DOSBox doesn't make it work all the options are going to involve a fair amount of work and a lot of overhead. The simple fact is that it is not trivial to maintain compatibility with the older architectures and there simply isn't enough demand for it. Most people end up in one of three camps,
