SO-50 has a very, very sensitive uplink. I've gotten into it with 50 mW to a whip antenna. Be sure to always use a 67.0 Hz tone.
The 250 mW downlink can be weak and you need high quality coax. For a home station, you almost certainly need a preamp unless you have a very short run of very good coax.
